Photoactivatible Janelia Fluor Dyes
Overview
Photoactivatable (PA) Janelia Fluor dyes combine small-molecule fluorophores' superior brightness, photostability, and cell permeability with precise light-controlled activation. By incorporating a diazoketone caging group onto Janelia Fluor scaffolds, these dyes allow for on-demand fluorescence activation, enabling high-resolution imaging at the single-molecule level.
Key Advantages
- On-Demand Activation Activation by short-wavelength light (e.g., 405 nm) enables precise temporal and spatial control for single-particle tracking (sptPALM) and PALM imaging.
- Exceptional Brightness After activation, dyes achieve photon outputs comparable to or superior to those of leading fluorescent proteins like mEos3.2, delivering enhanced localization precision.
- Live-Cell and Fixed-Cell Compatibility Small, membrane-permeant ligands are compatible with HaloTag and SNAP-tag labeling strategies.
- Multicolor Capability Distinct spectral properties of PA-JF549 (green-orange) and PA-JF646 (far-red) allow for simultaneous two-color imaging in live or fixed cells.
